From Thiruvananthapuram (Trivandrum), you can take a bus to Nedumangad. From Nedumangad, hire a jeep or taxi to reach Peppara Wildlife Sanctuary. The sanctuary is located near Ponmudi, so you can also take a bus to Ponmudi and then hire local transport.
